First of all, if you familiar with the original Dungeon Keeper, most of the strategies that worked in the original will work in DK2.

As I get time, I'll add more strategies, but here are a few basic ones to get you started.

When you're presented with a new realm to conquer, pay close attention to the narrative at the beginning.  This narrative provides important  information about what to expect, and vital clues about how to proceed.

Then, look at the map by pressing the "M" key.  While the Fog of War hides most of the details of the map, you can still get a pretty good idea of your current situation and where to expect trouble by your position on the map.

Plan your dungeon!  You need to create tight groups of basic rooms, so that creatures don't have to travel far between Lairs, Hatcheries,  Training rooms, and Treasuries.  In DK2, monsters seem to become unhappy even while traveling to the room they need, so the trips must be as short as possible!  Your Dungeon Heart serves as a treasury, so you need not build one near it, but you should have a treasure room in every far-flung "Mini Dungeon" you build.. 

In DK2, creature are easily annoyed, so you will need multiple sets of basic rooms to keep natural enemies apart.  Each "Mini-Dungeon" should have at least one Lair, Hatchery, training room, and treasure room.  When you "specialize" each Mini-Dungeon by adding rooms like Libraries, make sure that the creatures that use those rooms establish their Lairs in that Mini-Dungeon.

If possible, make sure there's only one way into your dungeon, and make sure that invaders have to pass thru at least one training room and lair to reach your Dungeon Heart.  Training rooms aren't as useful as they were in DK1, since only creature below level 4 will train there, but even these creatures can slow down enemies long enough for  you to bring in some reinforcements.

Use lots of traps to protect the way into your dungeon.  traps can slow down and wear down enemy forces before they start killing your creatures.
